Abstract
Objective To determine the effects of human mitochondria] transcription factor A (TFAM) on the cisplatin resistance of andro-gen independent cell line PC3. Methods Western blot was used to check the expressions of TFAM in the PC3 ceD line and in the cis-platin-resistant cell line PCDP6. After knocking down the TFAM in the PC3, the sensitivity to cisplatin was detected, and IC50 was calculated. Results Compared with its parent cell line PC3, the PCDP6 showed a higher expression of TFAM. The PC3 cells become highly sensitive to cisplatin after knocking down of TFAM by siRNA; at the same time, the expression of suivivin decreased. Conclusion The expression of TFAM improved the resistance to cisplatin in PC3 cell line,and this function of TFAM may be related with the survivin expression.关键词
