Effect of Peimine on Inhibiting the MCF- 7/TAM Proliferation of Human Breast Cancer Cell and Its Influence on Cell Apoptosis

Abstract
Objective; To investigate the inhibit proliferation and its mechanism of peimine on human breast cancer cells resistant to tamoxifen( MCF -7/TAM). Methods: Detect inhibitive rate under the influence of peimine on MCF -7/TAM cells by the method of tetrazolium blue( MTT) colorimetric assay, detect cell cycle,apoptotic rate by flow cytometry and detect changes in the expression of Bcl - 2 by immunocytochemistry. Results: Peimine had a significant inhibition on MCF-7/TAM cells, which depended on concentration and time. It could induce apoptosis after 48 hours. The cell cycle was arrested at Gl phase (P <0.05). It was detected by immunocytochemistry that the expression of Bel -2 decreased (P <0. 01). Conclusion; Peimine can inhibit the proliferation of MCF -7/TAM cells and induce its apoptosis.关键词
