Construction and evaluation of the segregation population in Malus sieversii

Abstract
[Objective]The objective of the study is to construct the optimal segregation population for constructing the genetic linkage map in Malus sieversii. [Method]Using red ‘Fuji' ×Xinjianghongrou apple as experimental material, a segregation population for M. sieversii had been established by artificial pollination, and the polymorphism between parents, hybrid purity and genetic structure among all progenies had been detected by microsatellite marker. [Result]The results indicated that there was larger genetic variation between red'Fuji' and Xinjianghongrou apple. Excluding 15 undefined progenies, 110 progenies of the segregation population were used to construct the genetic linkage map in M. sieversii. There was no repetition phenomenon among all progenies. 232 polymorphic fragments were detected by 64 SSR primers, among which 191 (82.3%) segregated in the expected Mendelian 1:1 ratio (P〈 0.01). [Conclusion] The studies suggested that the segregation population of red ‘Fuji' ×Xinjianghongrou apple was optimal to construct the genetic linkage map in M. sieversii.关键词
