外源赤霉素诱导‘藤稔’葡萄果实差异表达基因的cDNA—RAPD分析
Identification of differentially expressed genes by cDNA-RAPD approach during GA treatment in 'Fujiminori' grapevine fruits

Abstract
[Objective]The objective of this study is to identify genes in response to gibberellin treatment. [Method]cDNA-RAPD approach was employed to identify genes differentially expressed in ‘Fujiminori' grape fruits with the treatment of gibberellin. [Result]Sixteen random primers were used for selective amplification. Sixty-one TDFs were screened for their differential expression under gibberellin treatments. Among the 61 transcripts drived fragments (TDFs), 42 TDFs were up-regulated and 19 TDFs were down- regulated. 18 TDFs were successfully cloned and the sequences were analyzed. The results indicated that 17 TDFs had homology to known genes, and 10 of which displayed homology to genes with known functions. One TDF didn't match to known genes. Semi-quantitative RT-PCR analysis with 6 selected TDFs indicated that 3 genes all differentially expressed or up regulated in the fruits after treatment with GA, the other 3 TDFs did not express or down regulate in the fruits after treatment with GA. These results were similar to those of cDNA-RAPD expression patterns in grape fruits. [Conclusion]In this study, some differently expressed genes that were induced by gibberellins were investigated by cDNA-RAPD techniques.关键词
