桃果实类胡萝卜素测定方法的研究
Study the measurement of carotenoids from the peach fruit

Abstract
【Objective】The objective of the study was to explore the determination method and analysis the main composition and content of carotenoids in peach fruits.【Method】The carotenoids content of the mature yellow fruits of 'Independence' peach in was tested by UV spectrophotometer and High Performance Liquid Chromatoqraphy(HPLC).The different extraction solvent and method,chromatographic column,main composition and content of carotenoids were compared.【Result】The best condition of the determination method of carotenoids in peach fruits was screened out as follows: acetone as extraction solvent,directly extracted without saponification,and filtered after 0.22 mm organic filter head.Used YMC-C30 column,mobile phase A is Methanol,mobile phase B being Methyl tert-butyl ether and mobile phase C being super water.Flow rate is 1.0 ml·min-1;UV at 450 nm;the temperature being 25 ℃ and 100 min,sample introduction being 20 μL,through gradient elution with 90 min from 81%A/15%B/4%C to 6%A/90%B/4%C.The condition should be back to initial state after 92 min from 81%A/15%B/4%C.【Conclusion】 The method suggested here can be used in qualitative and quantitative analysis of carotenoids in peach fruits..关键词
