10-羟基喜树碱对类风湿关节炎患者Th17细胞功能的影响
Effect of 10-hydroxycamptothecin on Th17 cells of patient with rheumatoid arthritis
摘要
Abstract
Aim: To investigate the effect of 10-hydroxycamptothecin( HCPT ) on Thl7 cells in peripheral blood of patients with rheumatoid arthritis( RA ). Methods: Sixty active RA patients were enrolled and divided into three groups ( HCPT, MTX, and sham groups ),and PBMCs of the 3 groups were separated and cultured with HCPT, MTX, and normal saline for 48 h, respectively. PBMCs separated from 20 healthy persons and cultured with normal saline for 48 h were the normal control. The percentage of Thl7 cells was detected by intracelluar flow cytometry, and the level of IL-17 in the supernatant was tested by ELISA before and after the culture. Results: The percentage of Thl7 cell and IL-17 secretion level of RA patients were higher than those of the normal control( t = 47. 021,12. 507 ,P < 0. 001 ). The percentage of Thl7 cell and IL-17 secretion level decreased significantly in HCPT group and MTX group compared with the other 2 control groups( F = 418.743,142. 112,P<0.001 ). Conclusion: HCPT can inhibit the function of Th17 cells.关键词
