Abstract

The surfactin produced by Bacillus.amyloliquefaciens XZ-173 in liquid-state fermentation was optimized by Plackett-Burman and Box-Behnken design using response surface methodology.First,a Plackett-Burman design was used to evaluate the significant influence of the 17 factors related to the production of surfactin and MnSO4,CuSO4,temperature and rotational speed were found to be the significant.Then,the values of the four factors above were further optimized using a Box-Behnken design,and the optimum values of these factors were 4.4 mg/L of MnSO4,0.18 mg/L of CuSO4,temperature of 29℃ and rotational speed of 163 r/min.Results of the response surface analysis showed that the optimum production was obtained when the cells were grown on medium containing 15 g/L of glucose,5 g/L of peptone,6 g/L of L-monosodium glutamate,0.8 of g/L KH2PO4,0.4 g/L of MgSO4,0.4 g/L of KCl,3 g/L of yeast extract,2.5 mg/L of L-phenylalanine,4.4 mg/L of MnSO4,0.18 mg/L of CuSO4,0.12 mg/L of FeSO4,pH of 7.5.Meanwhile,the optimum cultivation conditions were temperature of 29℃,rotation rate of 163 r/min,inoculum quanlity of 5%,broth content of 50 mL in 250 mL,and cultivation of 36 h.The average yield of surfactin under optimized conditions reached 336 mg/L,and the actual experimental results were in agreement with those of prediction.The surfactin was extracted by HPLC,and its EI24 could reach as high as 70% at the concentration of 50 mg/L,indicating that it had excellent emulsifying property and potential application in industries.
