Abstract

Three magnetic fields(hypomagnetic fields,HMF;geomagnetic fields,GMF;moderate static magnetic fields,MSMF) and four culture mediums(NB1,NB2,MS1 and MS2) were applied to investigate the effects of magnetic field intensity and culture medium ingredient on callus induction in Arabidopsis(Columbia type).The results indicated that magnetic intensity and culture medium ingredient had different effects on callus induction in whole process.In the first 3~6 days after inoculation,the callus induction rate under HMF was inhibited,but the rate under MSMF was promoted significantly contrasting with the GMF condition.As far as the culture medium was concerned,in the first 3~7 days after inoculation,the callus induction rate on NB2 and MS2 medium were significantly higher than that on NB1 and MS1.Callus net weight reflects the state of cell proliferation in some extent.Compared to the GMF condition,there was no effect on cell proliferation under HMF condition,while the inhibition effect under MSMF was significant.Cell proliferation on NB2 and MS2 medium was increased more significantly than that on NB1 and MS1.All of these results showed that the callus induction rate in early stage and cell proliferation were affected significantly by magnetic field intensity and medium composition.However,there was no effect on the total callus induction rate after 15 days of inoculation,and no interaction between the two factors.Under HMF and GMF conditions,the callus status on NB2 and MS2,added 6-BA showed bright color,moderate density and useful for the subsequent subculture.
