PRELIMINARY STUDY OF IRRADIATION ON BIOLOGICAL EFFECTS OF GRAPE MATURE SEED

Abstract
The seeds of two varieties of grapes,Yongyou No.1 and Jufeng,were treated by 60Co γ-rays irradiation of 10,20,50 and 100Gy,respectively.The seedlings from treated seeds were used to detect the germination rate,reduced glutathione,superoxide dismutase,peroxidase,catalyse and seedling growth to investigate the effect of irradiation treatment on the growth and anti-oxidant enzymes of grape and develop the suitable dosage for grape seeds irradiation in mutation breeding of grape improvement.The results showed that the germination rate of augrape seeds were significantly decreased with LD50 of 33Gy.Leaf anti-oxidant enzyme activity assays showed that these four enzyme activities were firstly increased and then significantly decreased with the increase of irradiation dosage with both grape varieties,which may because that low-dosage irradiation can stimulate the antioxidant enzyme activities of seedling leaves of grape,while high dosage irradiation can inhibit these activities.In addition,the stem length of seedlings increased,with the dose(200Gy) increasing.Considering all above results,20~50Gy was recommended as a suitable irradiation dosage of grape seeds.关键词
