Abstract

Differentially expressed proteins were isolated by the two-dimensional (2D) gel electrophoresis, and analysed by the Image Master software, and identified by the mass spectrometry, after the extraction of the total protein of the control and the ethylene-induced early-ripening banana peel. A putative chitinase M (MaCH HI) was picked out for subsequent research from the differentially expressed proteins. The nucleic acid sequence to corresponding peptide sequence of MaCH HI was found by the NCBI BLAST. The full-length DNA sequence of MaCH IH gene was obtained by polymerase chain reaction (PCR) using PCR primers, which were designed according to the nucleic acid sequence. The results showed that this sequence share about 99% homology to a II class acidic chitinase in other species. And then, it was preliminarily confirmed as a gene of HI kind of acidic chitinase in banana.
