摘要

Abstract

To firstly report the isolation of quercetin 3-0-glucopyranoside and quercetin3-0-gentiobioside from the crude extract of okra, and establish an HPLC method for determination of the two flavonoid glycosides. Methods: Welch Materials XB-C18 (4.6 mm×250 mm,5μm) column was adopted; The mobile phase was methanol-water (42: 58) at the flow rate of 1 mL·min-1; The detecting wavelength was 256 nm, and the column temperature was 30℃. Results: The method had a good linearity in the range of 0. 165 μg ~ 0. 825μg (r = 0. 9999) for quercetin 3-0-glucopyranoside and 0. 155 μg ~ 0. 775 μg( r = 0. 9995 ) for quercetin3-0-gentiobioside. The recoveries were 92% and 100.3%. Conclusion; The method is simple, accurate and reproducible. It can be used for determination of flavonoid glycosides in okra.
