树突状细胞免疫应答功能的形态学证据及钙离子浓度变化的研究

Abstract
Objective To explore the morphological evidence of dendritic cells (DCs) inducing immune responses in vitro and the concentration of calcium of DCs once exerting antigen-presenting function for providing biochemical parameters of DCs functional researches and clinical treatment. Methods The mature DCs were induced in vitro. The morphological characteristic of DC, the expression and distribution of CD86 and Cdla and the interaction of DC with T lymphocytes were observed by light microscope, confocal microscopic system and transmission electron microscope. With Fluo-3 Ca2+ as the marker, the concentration of Ca2+ in all of DCs and in single DC was tested when the DCs were contacted with T lymphocytes. Results The DCs induced in vitro had the classical dendriti shapes and expressed abundant CD86 and Cdla on its membrane. Antigens of DCs were submitted rapidly and the concentration of Ca2+ was changed significantly in the all DCs and in single DC while DCs interacted with T lymphocytes. Conclusions The DCs induced in vitro have the classical shapes, and express CD86 and Cdla mainly on its membrane. DCs can form immune responses groups with contacting T lymphocytes rapidly. The change of the concentration of Ca2+ in DCs may relate to the immune response.关键词
