Expression of Notch1, Jagged1, p-Akt and NF-κB in human gastric cancer and its significance

Abstract
Objective To investigate the expressions of Notchl, Jaggedl, p-Akt and NF-kB in gastric cancer and their correlation with clinicopathological features and Helicobacter pybori (Hp) infection. Methods The expression of Notchl ,Jaggedl ,p Akt and NF-kB was detected by tissue microarray and immunohistochemistry in 60 cases of gastric cancer,60 cases of precancerous and 20 cases of normal gastric mucosa and Hp infection was tested by Warthin-Starry silver stain. Results The positive rates of Notchl, Jaggedl,p-Akt and NF-kB in gastric cancer tissues were 40.0% ,70.0% ,55.0% and 66.7% ,respectively,significantly different from those in precancerous tissues(81. 7% ,96. 7% ,30.0% and 36. 7% respectively,P <0.05) and those in normal gastric mucosa tissues (85.0% ,100.0% ,5. 0% and 0% .respectively,? < 0. 05). The infection rates of Hp in gastric cancer, precancerous and normal gastric mucosa tissues were 76. 7% ,93. 3% and 10. 0% , respectively, there was significant difference between the groups(P< 0.005). The expression of Notchl .Jaggedl and Hp infection was not correlated with the clinicopathological features. The expression of p-Akt and NF-kB was significantly correlated with tumor differentiation status and TNM ( P < 0. 005). Positive correlation was shown between Notchl and Jaggedl (r = 0. 460, P < 0. 05). Negative correlation was shown between Notchl and p-Akt or Notchl and NF-kB ( r = -0.287,r= -0. 361,P<0.05). Positive correlation was shown between p-Akt and NF-kB(t = 0.284,P<0.05). No correlation was shown between the expression of them and Hp infection ( r = - 0. 032, r = 0. 155 , r = 0. 055, r = 0. 028, P > 0. 05 ). Conclusions There are low expression of Notchl and Jaggedl and high expression of p-Akt and NF-kB in gastric cancer and all of them participate in the ccurrence and progress of gastric cancer, but all of them are not associated with Hp infection. The down-regulation of Notchl/ Jaggedl signaling pathway may promote the occurrence of gastric cancer though regulating the expression of p-Akt and NF-kB.关键词
