重庆地区载脂蛋白E基因多态性与阿尔茨海默病的关系研究
Relationship between Apolipoprotein E Polymorphism and Alzheimer′s Disease in Chongqing
摘要
Abstract
Objective To investigate the genotype distribution of apolipoprotein E (ApoE) in patients with Alzheimer's disease (AD) and healthy elderly and to discuss the relationship between AD and ApoE, thus predicting the likelihood of AD occurrence in crowd and offering basis for the prevention of AD. Methods 68 AD patients (AD group) admitted to the out-patient department of the First Affiliated Hospital of Chongqing Medical University from 2009 to 2010 and 72 healthy elderly (control group) underwent physical examination in our hospital were selected. PCR-RFLP was used to determine the ApoE genotypes of the two groups and the genotypes and alleles frequencies were also compared between the two groups. Results The frequency of ApoE locus was in genetic equilibrium in both groups after detected by Hardy-Weinberg equilibrium test (P>0. 05) . The frequencies of ApoE genotypes in the AD group were significantly different from that in the control group (P = 0. 0309) by using Fisher's exact probability method. Comparison of each genotype between the two groups showed that the frequencies of ApoE ε4/ε4 in the AD group were significantly different from that of the control group (P = 0. 025) . The frequencies of alleles between the two groups showed statistically significantly difference (X2 =9.11, P = 0. 0076) . Comparison of frequencies of each allele showed that the frequencies of ε3 and ε4 in AD group were significantly higher than that in the control group (P<0. 05 , OR =0. 57 ; P<0. 01, OR = 2. 98) . Conclusion ApoE{>4 allele is high-risk factor for Alzheimer's disease. ApoE genotypes test can be used to predict the likelihood of AD in the crowd and screen high-risk people in order to provide relevant prevention.关键词
