Sequencing and genetic variation analysis of whole genome of porcine reproductive and respiratory syndrome virus Myanmar strain

Abstract
The first strain of porcine reproductive and respiratory syndrome virus was isolated from a pig farm with PRRS syndrome in Myanmar Buddhist state in 2010 and it was designated as Myanmar strain for short.Gene fragments of Myanmar strain were amplified by RT-PCR and then cloned into the pEASY-T3 vector for sequencing.The sequencing showed that the genome of PRRSV Myanmar strain was 15 411 nt in length(excluding PloyA tail),containing 10 open reading frames(ORFs)with 189 nt in the 5′UTR and 151 nt in the 3′UTR.The genome of PRRSV Myanmar strain showed 99.3% and 89.5% homology,with US prototype(VR2332) and highly pathogenicity PRRSV prototype(NVDC-JXA1),respectively,and no amino acids deletion was exhibited in nonstructural protein 2 region.And it showed 54.6% homology with European prototype(LV),indicating that the PRRSV Myanmar strain belongs to the American genotype strain in genetic diversity.At the same time,phylogenetic analysis showed that the PRRSV Myanmar strain had closer relationship with PRRSV 01NP1.2 strain,RespMLV strain,BJ-4 strain and PL97-1 strain.关键词
