鸡法氏囊B淋巴细胞全长cDNA表达文库的构建
Construction of full-length cDNA expression library of B lymphoid cells in the bursa of Fabricius by SMART technology

Abstract
The mRNA was extracted from B lymphoid cells in the bursa of Fabricius and its quality was identified using aldehyde-agarose gel electrophoresis.The cDNA was synthesized with the SMART and LD-PCR methods.The products were then digested with SfiⅠ and linked into the pEXP-Lib plasmid to construct the full-length cDNA expression library.The capacity of primary library and amplified library were 1.98×105CFU and 2.35×109 CFU,respectively.The recombination ratio was about 100%,with inserts varied from 500 to 2 000 bp.Sequencing results of the 22 clones randomly selected from the library suggested that 20 clones shared more than 97% identity with the Gallus sequence and the other 2 were unknown.The library was therefore possessed high capacity and recombination ratio,which provided good foundation for the screening of IBDV receptor gene or other genes related to IBDV infection.关键词
