Abstract
Objective To establish a HPLC method for determining the contents of psoralen and isopsoralen in Shengfa Tincture. Methods The HPLC method was adopted to determine the contents of psoralen and isopsoralen in Shengfa Tincture. The chro-matographic column was Diamonsil C18 column (150 mmx4. 6 mm,5 |xm),with the column temperature of 35 癈 and acetonitrile -water(24:76) as the mobile phase. The flow rate was 1.0 mL/min and the detection wavelength was 246 nm. The number of theoretical plates calculated according to psoralen peak was not less than 3 000. Results In the linear range of 12.04-301.0 (xg/mL for psoralen, its concentration and peak area showed good linear relationship; and in the linear range of 9.16-229.0 u,g/mL for isopsoralen, its concentration and peak area showed good linear relationship, both Methods conformed the technical requirements. The psoralen sample recovery rate was 99. 06% , RSD = 1. 34% ( n = 6) , and isopsoralen sample recovery rate was 98. 29% , RSD = 1. 86% ( n = 6). Conclusion The used method has good separation effect, sensitive, accurate, better reproducible, and can be used for the quality control of Shengfa Tincture.关键词
