livinβ对胶质瘤细胞增殖和凋亡能力的影响
Effects of livinβ gene disruption on apoptosis and proliferation of glioma cells
摘要
Abstract
Objective To investigate whether silencing livinp gene by siRNA leads to apoptosis,arrest of cell cycle and inhibition of proliferation. Methods The recombinant plasmid was transfected into U251 glioma cells by liposome mediated transfection. Then PI flow cytometric analysis was performed for cell cycle and apoptosis,MTT assay for cell proliferation,RT PCR and Western blot ting for caspase 3 expression. Results Silencing livinp gene by siRNA could inhibit cell proliferation remarkably(P<0. 05) ,could arrest cell cycle significantly and induce cell apoptosis(P<0. 05) ,could upregulate caspase 3 obviously(P<0. 05). Conclusion Si lencing living gene in glioma cells could activate caspase 3 and increased poly(ADP ribose) polymerase cleavage,therefore induce apoptosis and inhibit proliferation. So the livinp may play key role in pathogenesis of glioma.关键词
