Advancement of regulation of p66Shc expression and its role in diabetic nephropathy

Abstract
p66Shc, an adaptor protein, is known as a key regulator of cellular oxidative stress and life cycle, eventually leading to cellular oxidative injury. Diabetic nephropathy is a major cause of end-stage renal disease. Over-production of reactive oxygen species is regarded as the key process in the pathogenesis of diabetic nephropathy. In this review, we summarize the structure, function, transcription, post-translational modification to the p66Shc protein, and mitochondrial dysfunction of p66Shc gene, as well as the physiological relationships between mitochondrial dysfunction of p66Shc and reactive oxygen species, and the role of p66Shc in the pathogenesis of diabetic nephropathy.关键词
