石栗树腋芽外植体消毒方法的筛选
Screening of sterilization methods for explants of Aleurites moluccana axillary bud
摘要
Abstract
[Objective]Appropriate sterilization methods for explants of Aleurites moluccana axillary bud was selected in order to solve the pollution problem in tissue culture of Aleurites moluccna and improve survival rate of tissue-cultured Aleurites moluccana. [Method]Three factors (tap water flushing,0.1% HgCl2 soaking,and 10% NaClO3 soaking) and three levels of (60,120,and 180 min for tap water flushing; 0,8,16 min for 0.1% HgCl2 soaking; 0,12,24 min for 10% NaClO3 soaking) orthogonal test was designed to observe the sterilization effect on appearance of Aleurites moluccana branch processed by nine treatments. [ Result ]The results showed that the best combination for explants sterilization was as follows: flushed by tap water for 120 min,soaked in 0.l%HgCl2 for 16 min,and soaked in 10%NaClO3 for 24 min. [Conclusion]The above-mentioned best combination could bring down the contamination rate to 16.67% with no serious harm to explants. Explants obtained from treelet in low-temperature and dry season had little pollution to the environment.关键词
