Abstract

β - glucosidase was immobilized on chitosan beads by cross - linking and adsorption. The effects of pH, temperature on glycitin transformation reaction by immobilizedβ - glucosidase, the immobilized enzyme in repeated batches and its storage stability were investigated. The results showed that three glucosides of soybean isoflavone could be transformed by β - glucosidase. The optimal pH value was 6.4 and the optimal temperature was 45 ℃ for transformation of glycitin. The enzymatic activity of immobilized β - glucosidase remained more than 85% after stored at pH 6.4, 4 ℃ for 25 days. The yield of glycitein transformed by immobilized recombinant β - glucosidase kept about 85% after 10 batches.
