Expression of Gene pLXSN-MICA*008 and pLXSN-MICA*001 in Human Foreskin Fibroblast Cell
摘要
Abstract
Objective: Construct human foreskin fibroblast ( HFF) cell expression protein MICA * 008, MICA * 001 to provide biotic material to research immune function of gene MICA. Methods: Cell culture in vitro technique was used; The retroviral vector pLXSN, pLXSN-MICA * 008 and pLXSN-MICA * 001 transfect packaging cell PA317 with lipo-somes to gain virus particles. HFF cells are infected by virus particles and screened with G418 resistance for 14 days. The expression protein MICA * 008,MICA *001 were dentified by westernblot. Results:Packaged by packaging cell,the retroviral vector pLXSN, pLXSN-MICA * 008 and pLXSN-MICA * 001 form virus particles with infection ability which are able to infect HFF cell and protein MICA * 008, MICA * 001 was expressed in HFF cell indicated by westemblot. Con-elusion: HFF cell expression protein MICA * 008 , MICA * 001 was constructed successfully.关键词
