Effect of pretreatment with scutellaria baicalensis stem-leaf total flavonoid on morphologic changes of hippocampus neurons and blood brain barrier followed by focal cerebral ischemia reperfusion in rats

Abstract
Objective:To study the influence of scutellaria baicalensis stem-leaf total flavonoid (SSTF) on the morphologic changes of hippocampus neurons and blood brain barrier (BBB) induced by focal cerebral ischemia reperfusion in rats. Methods:50 SD rats were randomly divided into five groups: sham operation group, ischemia reperfusion group. SSTF pretreatment groups (SSTF 50 mg o kg-1 , 100 mg o kg-1 and 200 mg o kg-1 , respectively). The model of focal ischemia reperfusion in rats was established with suture occluded method invented by Zea Longa and improved properly in our lab. The changes of the neurological function, neuron density in hippocampus CA1 were measured 24 hours after cerebral ischemia reperfusion. The ultrastructure changes of hippocampus neurons and BBB in each group were observed under a transmission electron microscope. Results:SSTF pretreatment improved rats neurological behavior. Compared with I/R group, SSTF markedly induced the increase of survival pyramidal cells in the hippocampus CAl region, significantly improved the morphologic changes of hippocampus neurons and BBB and presented a dose dependence effective relations. Conclusion :SSTF pretreatment has an obviously protective effect on the morphologic changes of hippocampus neurons and BBB induced by focal cerebral ischemia in rats.关键词
