Abstract

Objective:To explore the feasibility that myocardial cells induced from insulin-like growth factor 1 (IGF-1) expressing embryonic stem (ES) cells to treat rat myocardial infraction. Methods: The recombinant containing cDNA segment of human IGF-1 was packed in PA317 cells mediated by Lipofectamine 2000, and transfected into ES cells. After the expression of IGF-1 was analyzed, the ES cells were induced into myocardial cells. And then, rat acute infracted myocardium was treated by implantation of myocardial cells differentiated from IGF-1 expressing ES cells used as IGF-1 group, or myocardial cells differentiated from ES cells used as myocardial cells group, or DMEM medium used as control. Results: The myocardial cells induced from IGF-1 expressing ES cells showed upregulated expression of human IGF-1, compared to those in control. Immunohistochemical analysis indicated that implanted myocardial cells survived in infracted myocardium, and then, caused more neovasculature. Conclusion: The myocardial cells containing high expressing IGF-1 can promote the formation of neovessels, therefore, is benefit to survival of implanted cells, and improve the cardiac function.
