摘要

Abstract

High pathogenicity island (HP1) is first detected in Yer.sinia. It was found in many kinds of Escherichia coli. The irp1 gene and fyuA gene were analysed by DNAStar to look for the surface probability plot. Eight fragments of irp1 gene and two of fyuA gene were selected. Primers were designed to amplify the ten fragments. The PCR products were cloned into expressive vector pET32a and transformed into fimbriae negative host bacteria BL21. The recombinant bacteria were induced with IPTG to expression and mixed with the single layer Vero cells growing well in dishes. After washed six times with Hank's solution, the cells were observed with oil microscope. The results showed that the recombinant bacteria BL21-rpET32a-irp1-1 including irp1-1 fragment had strong reaction with Vero cells. The production irp1-1 gene encoded may act as an bridge between the bacteria and host cells during process of pathogenicity.
