Abstract

Objective To study the effects of daidzein on proliferation and osteogenic differentiation of stem cells derived from human exfoliated deciduous teeth ( SHED). Methods The third passage SHEDs were incubated with 0.1, 1, 10, 100 |xM daidzein, then cells proliferation curves were examined by MTT method. Alkaline phosphatase (ALP) , osteocalcin (OCN) and TGF- (3 1 were detected by alkaline phosphatase kit, radio-immunity method and TGF- f$ 1 Elisa kit, respectively. Results 0.1 (xM daidzein improved proliferation of SHED, 100 (xM daidzein, however, showed a depressant effect on proliferation. ALP, OCN and TGF-|5 1 expression were enhanced by daidzein in a dose-dependent manner. Conclusion Daidzein enhances proliferation and osteogenic differentiation of SHED, and the mechanism may be related with upregulating TGF?{$ 1 expression.
