青钱柳幼胚愈伤组织的诱导
Callus induction from young embryos of Cyclocarya paliurus
摘要
Abstract
Using young embryos of Cyclocarya paliurus as explants, we investigated the effects of embryo development period, basic media, types and the concentrations of plant growth regulators on callus induction of C. paliurus. The results showed that there were obvious differences in the shape of callus among different embryo development periods, and the middle cotyledon embryos were most suitable for callus induction. As a basic media, improved DKW medium was the best selection for young embryo callus induction, of which induction rate could hit to 88. 89 % with no browning phenomenon. The medium containing 2, 4 - D and KT was more appropriate for callus induction. The 2, 4 - D (0. 5 mg/L) + KT (2.0 mg/L) as well as 2,4 - D (1.0 mg/L) + KT (1. 0 mg/L) were optimal for callus induction of C. paliurus young embryo, and the induction rates were 80.00 % and 78.33 % respectively.关键词
