缺氧后处置心肌细胞CYP2J3/EETs变化及PPOH对其的影响
CYP2J3/EETs changes of primary myocardial cells in hypoxia postconditioning and the effect of PPOH on it

Abstract
Objective To observe the CYP2J3/EETs changes of primary myocardial cells in hypoxia postconditioning and the effect of PPOH of epoxygenase inhibitor on it. Methods Primary cardiac cells from Wistar neonatal rats( 12-24 h) were cultured,and randomly divided into five groups as follow; control( CON) group, hypoxia/reoxygenation( H/R) group, hypoxia postconditioning ( HPost) group, DMSO solvent group, PPOH inhibitor group. Myocardial activity was tested by MTT, the expression of CYP2J3 was assayed by Western Blotting, and 11,12-EETs density of cell culture medium was tested by HPLC. Results The activity of myocardial cell, the expression of CYP2J3 and the density of 11,12-EETs in H/R group were significantly lower than those in CON group, which in HPost group were significantly higher than those in H/R group, the activity of myocardial cell, the expression of CYP2J3 and the density of 11,12-EETs in PPOH group were lower than those in HPost group. Conclusions Myocardial cells can be protected by ischemic postconditioning through increase of CYP2J3/EETs, PPOH can weaken the protection of ischemic postconditioning by inhibitting the expression of CYP2J3.关键词
