Abstract
Gouty arthritis is one of the most painful symptoms of gout patients. This research aimed to investigate the effect of several natural extracts on acute gouty arthritis in rats induced by monosodium urate (MSU) crystals. The animal model of acute gouty arthritis on wistar rats wag made through injecting MSU in ankle joints. The rats were administrated orally with Fucoidan,Potassium Citrate and Tongkat Ali extracts. And the joint swelling,gait score and several inflammatory factors in serum were measured to discuss the prevention and relief effects in the inflammation-related limb swelling,pain,dysfunction of the natural extracts on acute gouty arthritis and investigated the mechanism preliminarily. As a result, the three kinds of natural extracts could significantly reduce the joint swelling after induction MSU in rats 24 h later. This showed that the three could inhibit the symptoms inflammation-related limb joint swelling. Tongkat Ali extracts could decrease the gait score. It means that the limb inflammation,pain and dysfunction had been reduced on the rats. Anti-inflammatory mechanisms of the three active substances were explored,by further testing of experimental rat serum biochemical indices. Fucoida and Tongkat Ali extracts could significantly reduce the level of Myeloperoxidase. It indicated that both the mitigation of the inflammatory response may be related to the activation of neutrophils. And Tongkat Ali extracts could significantly decrease the IL-1β,IL-8,NO and 5-HT of model rats. It showed that Tongkat Ali extracts had a certain inhibitory effect on the occurrence,development and other aspects of inflammation, while the symptoms of limb dysfunction was reduced by inhibiting the pain related inflammatory factor such as 5-HT. In conclusion,the three kinds of natural extracts can relieve the attack of acute gouty arthritis induced MSU in rats. And the Tongkat Ali extracts play an important role in reducing the joint swelling, relieving the pain and the inhibition of inflammation.
