Abstract

Objective To investigate the effect of silencing Notchl gene on cell proliferation in vitro in gastric cancer cell line SGC-7901 and the underlying mechanisms. Methods The siRNA of notchl was transfected into gastric cancer cell line SGC-7901 by Lipofectamine 2000. Proliferation and invasion of SGC-7901 were detected after transfection by methabenzthiazuron assay and transwell assay, respectively. The mRNA expression of Notchl was determined by RT-PCR. The protein expressions of Cyclin Dl, Cyclin Al and CDK2 were determined by Western blotting. The mRNA expressions of MMP-2 and COX-2 were determined by RT-PCR. Results Compared with that in control group, the mRNA expression of Notchl was significantly decreased after transfection. The proliferation and invasion of SGC-7901 cells were inhibited significantly after Notchl silencing. Additionally, the protein expressions of Cyclin Dl and Cyclin Al as well as the mRNA expressions of MMP-2 and COX-2 were obviously inhibited. However, silence targeting Notchl did not affect CDK2. Conclusion RNA silence targeting Notchl can inhibit the proliferation and invasion of gastric cell line SGC-7901 probably through decreasing the expressions of Cyclin Dl and Cyclin Al and down-regulating the expressions of MMP-2 and COX-2, respectively.
