摘要

Abstract

Objective To observe the effect of globular domain of adi-ponectin on glucose metabolism and its mechanism in 3T3 - LI adipo-cytes model which have generated insuline resistance ( IR). Methods Culturing and differentiating 3T3 - LI adipocyte. IR model of 3T3 - LI adipocytes were prepared with palmitic acid. Intervention the adipocytes which have been generated IR with different concentrations of gAd. The cell culture medium glucose content was detected with the glucose oxidize method. The mRNA expression of AMPK, CPT - I were detected with real - time quantitative PCR method. The phosphorylation of AMPK Thr -172 was detected by Western blot. Results Gad can promote glucose uptake of 3T3 - LI adipocytes model which have generated IR. Different concentrations of gAd gradually increased the mRNA expression levels of AMPK, CPT - I and in a dose - dependent manner. The AMPK Thr -172 phosphorylation levels increased gradually followed by the concentration of gAd ( P = 0. 000 ). Conclusion gAd can improve insulin resistance by AMPK signal transduction in 3T3 - LI adipocytes model which have generated IR.
