Abstract
Objective To investigate the effects of Angelicanaphtha on proliferation cell cycle, apoptosis, and collagen synthesis of human hypertraphic scar fibroblast. Methods Human hypertrophic scar fibroblast was cultured and passaged in Dulbecco's modified Eagle's medium (DMEM) and treated with Angelicanaphtha 1mg/L, 4mg/L, and 16mg/L at 1, 3, 5, and 7day respectively. The proliferation was measured with MTT method. The cell cycle and apoptosis were analyzed with Flow cytometry and collagen synthesis was determined with radioimmunoassay. Results Angelicanaphtha of 1mg/L, 4mg/L, and 16mg/L made G0/G1 phase cells, G2/ M phase cells decrease and S phase cells remarkable increase (P<0.05). Furthermore, apoptosis of the fibroblasts was increased with increase of Angelicanaphtha concentration (P<0.05). The collagen synthesis of fibroblast was inhibited by Angelicanaphtha in concentration-dependent manner (P<0.05). Conclusion The proliferation and collagen synthesis of fibroblast was inhibited by Angelicanaphtha.关键词
