Abstract
Objective To investigate the expression role of miRNA - 148b in breast cancer. Methods 80 samples of breast cancer tissues from breast cancer patients admitted to our hospital from March 2008 to March 2010, 80 samples of breast cancer adjacent tissues and 20 samples normal breast tissues were selected. The miRNA - 148b level was detected by real - time PCR technology. Three groups were divided when cultivating breast cancer cells: the blank control group was not given any medicines ; the negative control group was given placebo; the inhibitor group was treated by 10 μg/ml 5 - fluorouracil for 48 h, and then MDA - MB -231 cells were cultivated and transfected. Results The miRNA - 148b levels between normal breast tissues, breast cancer adjacent tissues and breast cancer tissues showed statistically significant differences ( P < 0. 001 ) . The miRNA -148b level in breast cancer tissues was significantly lower than that in the normal breast tissues and breast cancer adjacent tissues ( P <0. 05 ) . After 24 h transfection, the miRNA - 148b levels in blank control group, negative control group and inhibitor group showed statistically significant differences ( P < 0. 001 ) . The miRNA - 148b level in the inhibitor group was significantly higher than that in the blank control group and negative control group ( P < 0. 05 ) . After 24 h transfection, the proliferation of MDA - MB - 231 cells between the three groups showed no statistically significant difference ( P > 0. 05 ) . After 48 h and 72 h transfection, the proliferation of MDA - MB - 231 cells showed statistically significant differences between the three groups ( P < 0. 01 ). After 48 h and 72 h transfection, the proliferation of MDA - MB - 231 cells in the inhibitor group was significantly lower than that in the blank control group and negative control group ( P < 0. 01 ) . Conclusion The expression of miRNA - 148b in breast cancer tissues and MDA - MB - 231 cell strains is lower than that in normal tissues and breast cancer adjacent tissues, besides , the high expression is accompanied by increased cell proliferation. After transfection of inhibitor of miRNA - 148b, the expression of miRNA - 148b increases and cell proliferation decreases, indicating that miRNA - 148b may play an important role in the occurrence and development of breast cancer.关键词
