Flotillin-1缺失突变体基因酵母双杂交诱饵载体的构建与鉴定
Construction and identification of flotillin-1 deletion mutant bait vector in yeast two-hybrid system and detection of its self-activation and toxic effect

Abstract
To construct flotillin-1 bait vector pGBKT7-X for the screening oi priori protein interaction molecular area,the flotillin-1 gene fragments were amplified by RT-PCR from bovine brain tissue, and cloned into the the pGBKT7 vector. The recombinant vectors were transformed into AH109,with the self-activation and toxic action of pGBKT7-X tested. Results indicated that parts of pG- BKT7-X were successfully constructed and proven to be no self-activation and no toxic to the host yeast. Two of pGBKT7-X proven to be self-activation and no toxic to the host yeast. The parts of pGBKT7-X could be used as bait to screen the protein molecular area interacting with prion protein in the yeast two-hybrid system.关键词
