Abstract

OBJECTIVE: To develop a method for the content determination of dihydromyricetin (DMY) and myricetin (MY) in leaves of Ampelopsis grossedentata, and to investigate the stability of DMY and MY in leaves of A grossedentata. METHODS: UPLC method was adopted. The separation was performed on Dionex Acclaim (R)RSLC PA2(30 mm×2.1 mm, 2.2 μm) column with 0.1% phorsphoric acid-acetonitrile as mobile phase (78:22, V/V) at flow rate of 0.4 mL·min-1. The detection wavelength of DMY was set at 290 nm (0~2 min), and that of MY was set at 373 nm (2~4 min). The changes of DMY and MY content were detected by hot reflux extraction. RESULTS: The linear range was 200.0~1 600.0 ng(r=0.999 8) for DMY and 5.0~56.2 ng(r=0.999 7) for MY with average recovery of 98.74% (RSD=1.90% , n=9) and 99.78% (RSD=1.37% , n=9). The content of DMY was 0.53%~35.01% and that of 0~0.62% , and total content of them was 0.53%~35.30%. DMY had poor thermal stability, so it was not recommended to use hot reflux for extraction. CONCLUSION: The method is fast, accurate and helpful for the quality control of leaves of A. grossedentata.
