Abstract
Objective To study the effect of small interfering RNA (siRNA) mediated VCC-1 silencing on proliferation of hepatocellular carcinoma cell line SMMC7721. Methods Recombinant interfering plasmid was transfected into hepatocellular carcinoma cell line SMMC7721 by lipofectamineTM 2000. The efficiency of VCC-1 down regulation was assessed by Western blot. The effects of VCC-1 down regulation on the proliferation of SMMC7721 cells were determined by MTT assay and plate clone formation assay. Results Compared with the negative control and parental group,Western blot results demonstrated that the protein level of VCC-1 was significantly decreased (P < 0.01). Silencing of VCC-1 in SMMC7721 cells significantly suppress cellular proliferation rate and inhibit the ability of colony formation (P < 0.05). Conclusion Targeting of VCC-1 gene by siRNA can significantly inhibit the growth and proliferation in SMMC7721 cells.关键词
