摘要

Abstract

Aim To study the effeet induced by three cyclic hexadepsipeptides isolated from fungi and to explore its mechanism. Method Human PC-3 cells were cukured in vitro and then treated with cyclic hexadepsipept ides at the concentration of 10 μmol · L~1 . The effeet of three cyclic hexadepsipeptides on human PC-3 cells was measured by MTT assay. The apopLosis of the cells was evaluated by TUMEL assay. The protein expression of apopLosis in PC-3 cells was analyzed by WesLern blot assay. Results After three cyclic hexadepsipeptides ( 10 μmol ·L~1 ) LreaLment, Pullularin C showed significant activities against human PC-3 cells; Cell apopLosis of Pullularin C treated group was significantly higher than that in control group, and which also significantly up-regulated the Bax and caspase-3 protein expression compared with control group in PC-3 cells. Conclusions Pullularin C has a significanL inhibiLory effcct on the protiferation of human PC-3 cells. Besides, Pullularin C can also induce cells apoptosis through the mediation of Bax and caspase-3.
