大黄素对鼻咽癌CNE-1细胞放射增敏作用与细胞自噬关系的研究
Study of the relationship between the radiotherapy sensitization function of emodin and autophagy in the nasopharyngeal carcinoma CNE- 1 cells

Abstract
Aim To invesLigaLe vvheLher emodin induces autophagy in Lhe nasopharyngeal carcinoma CME-1 colls aftcr radiaLion combined with emodin and explore the relationship between the radiotherapy sensi-tization and autophagy-related gene mRMA expression. Methods Masopharyngeal carcinoma CME-1 cells were divided into control group, emodin alone group, radiation alone group and radiaLion combined with emodin group. After different treatmenLs, the ultrastructur-al changes of autophagosome were observed by trans-mission electron microscope( TEM ). The mRMA expressions of ULK.1 , GABARAPL1 , Beclinl and Bcl-2 were detected by Lhe Real-time PCR. Results Autophagosome or autophagolysosome was observed in all groups except in the control cells. Compared with the control group, the resuks of Real-time PCR showed that ULK1 mRMA expression in all the experimental groups increased( P <0. 05 ). The radiotherapy sensiti-zation group showed the highest( P <0. 01 ). However, Beclinl and Bcl-2 mRMA expression were down-regu-lated. GABARAPL1 expression had no remarkable changes among the experimental groups, exeept in radiotherapy sensitizaLion group where it was up-regulated ( P <0. 05 ). Conclusions Emodin induces autophagy in the nasopharyngeal carcinoma CME-1 cells after radiation combined with emodin. Autophagic cell death ( ACD ) may be one of the paths of the radiosensitiza-tion sensitization effect.关键词
