Effects of polysaccharides from Dendrobium Officinale on the production of TNF-α by RAW264.7 cells
摘要
Abstract
Aim To sLudy die effects of polysaccharides from dendrobium officinale ( DOP ) on the pro-due don of TMF-a in RAW 264. 7 cells and its possible mechanism. Methods Cell proliferation was measured by MTT assay; Concentration of TMF-a in super-natants was determined by FLISA kit; Expression of TMF-α mRMA was measured by reverse transcription polymerase chain reaction ( RT-PCR ); Production of 1-κBα protein was detected by Western blot. Results DOP (20-160 mg o L-1 ) possessed the stimulation effect on macrophages proliferation without cytotoxic. The production of TMF-α was increased in a dose-dependent manner when pretreated with DOP. Compared with control group, expression of TMF-α mRMA was significantly up-regulated. DOP markedly decreased intracellular 1-κBα level as demonstrated by Western blot assay. Conclusion DOP induces TMF-α gene expression followed by TMF-α production in macropha-ges via decreasing intracellular 1-κBα level, activating transcription factor, MF-ΚB, suggesting that the immu-noregulation mechanism of DOP may be related to the induction of TMF-α production in macrophages.关键词
