革皮氏海参总皂苷对小鼠骨髓细胞周期和细胞凋亡的影响
Effects of saponin of pearsonothuria graeffei on bone marrow cells in myelosuppression mice induced by cyclophosphamide

Abstract
Aim To investigate the effects of saponin of pearsonothuria graeffei ( SP )on coll cycle and apoptosis of hone marrow cells in cyclophosphamide( CY )-induced myelosuppression mice. Methods The myelosuppression mice model was generaLed after injected in-traperitoneally with CY( 100 mg · kg-1 ). The myelosuppression mice were divided into three groups: the model control group and saponin group treated with SP 10mg/( kg · d ) respectively. The other mice were given with 0. 5% CMC solution as placebo. All agents were administered once a day for 16 d. Changes in cell cycle and apoptosis of hone marow cells were detected with FCM ( flow cylometry ). The expressions of cy-clinBl ,bcl-2,bcl-xl were determined with reverse tran-scription PCR( RT-PCR ). Results Saponin of pear-sonothuria graeffei significantly elevated hone marrow nucleated cell( BMNC ) and promoted the cell cyclc from S phase to G2/M phase, the expression of cy-clinBl was also increased in saponin group. The apop-tosis rate in myelosuppression mice was significantly reduced and the expression levels of bo 1-2 and bcl-xl were olovatod in saponin group compared vvith the mod- el control mice. Conclusion Saponin of pearsonothu-ria graeffei is beneficial for improving bone marrow cells division and proliferation, inhibking cell apopto-sis, recovering the function of hematopoiesis.关键词
