摘要

Abstract

Aim To investigate the function of primary culture of rat distal pulmonary arterial smooth muscle colls ( PASMC ) for in vitro studics of the pathophysiol-ogy and medicine intervcntion of pulmonary circulation diseases, including hypoxic pulmonary hypertension. Methods Microsurgery operations and collagenase di-gesLion were performed to isolate and cukure rat distal PASMC. 'Ihe rat distal PASMC were identified by morphological acLivity and immunofluorescence of smooth muscle a-acLin. Furthermore, the response of intracel-lular Ca2+ concentration ([ Ca2+ ]; ) of PASMC to 60 mmol o L-1 KC1 was tested by fluorescence microscope. Results The rat distal PASMC displayed a typical peak and vallcy growth pattern and smooth muscle a-actin showed a positive result. In addition, PASMC showed a clear increase in [ Ca2+ ]; in response to 60 mmol ·L-1 KC1 and nifedipine given at a 5 μmol o L-1 concentration completely blocked the [ Ca2+ ], response to KC1 in PASMC. Conclusion The rat distal PASMC show the morphological and im-munological characteristics of vascular smooth muscle cells and possess the voltage-dependent Ca2+ channels ( VDCC ) with normal functions. PASMC can be used to further explore the pathophysiological mechanisms and medicine intervention of pulmonary circulation diseases, including hypoxic pulmonary hypertension.
