Abstract
Objective To investigate whether hypoxia can induce epithelial-mesenchymal transition in gastric cancer cells SGC-7901. Methods After SGC-7901 cells were cultured under hypoxic conditions for different hours ( 0, 6, 12, 24,48 h ), cell morphological changes were observed. The expression of HIF-2α, E-cadherin, N-cadherin, Vimentin were detected by Western blot. The scratch wound assay was used to assess the invasion and migration levels of SGC-7901 cells. Results After exposure to hypoxia for 48 h, SGC-7901 cells changed gradually into the shape of fusiform and generated pseudopodium. The expression of HIF-2α, N-cadherin, Vimentin were up-regulated ( P < 0. 05 )and E-cadherin was significantly down-regulated( P < 0. 05 ). The invasion and migration levels of hypoxi-a-treated cells were increased as compared with those of control group. Conclusion Hypoxia can increase the likelihood of the epithelial-mesenchymal transition in gastric cancer cells and improve their invasion and migration levels.关键词
