Effects of Rhizoma Paridis Total Saponins on Growth of Gastric Carcinoma Cell Lines MNK-45 and MGC80-3

Abstract
To observe the effects of Rhizoma Paridis total saponins (RPTS) on the growth of human gastric carcinoma cell lines MNK-45 and MGC80-3 . Methods MNK-45 and MGC80-3 cells were treated with 5 , 10 , 20 , 40 , and 80 μg/ml RPTS for 24 , 48, and 72 h, and then methyl thiazolyl tetrazolium (MTT ) assay was used to measure the growth inhibition rates of the two cells . MNK-45 and MGC80-3 cells were treated with 10 jug/ml RPTS for 24 h , and then acridine orange/ethidium (AO/EB) staining was used to observe the morphologi -cal changes of the two cells . MNK-45 and MGC80-3 cells were treated with 10 and 20 jug/ml RPTS , and then flow cytometry was used to determine the changes in cell cycles of the two cells . Results RPTS (5-80 jug/ml) had significant inhibitory effects on the growth of MNK-45 and MGC80-3 cells (P<0 .05 or P<0 .01), and the higher the dosage was , the longer and stronger the inhibitory effect was (P<0 .01) . RPTS had significantly stronger inhibitory effects on MNN-45 cells than on MGC80-3 cells when applied at the same concentration for the same peri -od (P< 0.01). Both cells showed apoptotic features under a fluorescence microscope after being treated with RPTS. Treated with RPTS, MNK-45 cells were arrested in Go /Gi phase, and MGC80-3 cells in S phase. Conclusion RPTS can inhibit cell growth , induce apoptosis , and affect cell cycle in human gastric carcinoma cell lines MNK-45 and MGC80-3 , but the effects on cell growth and cell cycle were different in them .关键词
