The role and significance of Notch signaling pathway in the proliferation and apoptosis of H22 cells in mice

Abstract
Objective To observe the role of Notch signaling pathway in the proliferation and apoptosis of mouse hepatocellular carcinoma cell line H22, and to explore its possible mechanism. Methods Cultured H22 cells were randomly divided into A, B and C group, Notch signaling activator rhNF-KB, Notch signaling inhibitor DAFT, and PBS, were added separately into above three groups, then cultured together for 48 h. Cell proliferation was observed with MTT method (cell absorbance value), cell apoptosis was detected with flow cytometry method, and the treated cells were analyzed with immunohistochemistry method (fluorescence staining). Notch1, Hes1, and Bcll-2 mRNA and the corresponding proteins in cultured cells were detected with RT-PCR and Western blotting methods, respectively. Results After 24, 48, 72 and 96 h of cell culture, the difference of cell proliferation among A, B, C group was significant (all P <0.05). The apoptosis rates of A, B, C group were 25.52% ± 3.13%, 0.91% ± 0.18%, 8.91% ± 1.50%, respectively, the difference among three groups was significant (all P<0.05). Compared with group C, Notch1, Hes1, Bcl~2 mRNA and their protein expression in group A were significantly increased (P<0.05), whereas the Notch1, Hes1, Bcl~2 mRNA and their protein expression in group B were significantly decreased (P<0.05). Conclusion Notch signaling pathway plays an important role in the inhibition of cell proliferation and the promotion of cell apoptosis of mouse hepatocellular carcinoma cell line H22. The possible mechanism may be related to its regulation of the expression of target gene Hes1 and Bcl-2.关键词
