摘要

Abstract

Production of lipase by immobilized cells from Rhizopus arrhizus was studied in a horizontal bioreactor with polyurethane foam as a porous biomass support. The effects of immobilized areas, aeration rate, rotating rate and volume of broth on the lipase activity were investigated. The optimum fermentation conditions obtained were as follows; aeration rate 5 L/min, rotation speed 40 r/min, and 3. 5 L medium with the cylindrical bracket in 6 L horizontal bioreactor. The highest lipase activity of 95 U/mL and the lipase productivity of 2021 U/(h · L) were achieved in batch fermentation under the optimized fermentation conditions. Three repeated batch fermentation were carried out in 6 L horizontal bioreactor. The highest lipase activity of 110 U/mL and the average lipase activity of 95 U/mL were achieved. The lipase productivity increased from 2021 U/(h · L) in batch fermentation to 3476 U/(h · L) in repeated batch fermentation, which was 1. 72 times as high as that in batch fermentation. It was found that immobilized cells showed high stability for repeated use.
