Abstract
Objective To investigate the effects of Edaravone on brain water content, brain tumor necrosis faclor-α (TNF-α) content, serum level of matrix metalloproteinase-9 (MMP-9) and activity of superoxide dismutase (SOD) in rats with intracerebral hemorrhage (ICH). Methods Thirty-six SD rats were randomly divided into sham operation group, Edaravone-treated group and ICH group, with 12 rats in each group. ICH model rats were established by stereotactic infusing of autologous. Edaravone-treated group was treated with 3 mg/kg Edaravone by intraperitoneal injection, one time each 12 h untill death. Seventy-two hours later , brain contents of water was detected with wet and dry method, TNF-a and MMP-9 were detected with enzyme-linked immunosorbent assay (ELISA) respectively, and the activity of SOD was detected by xanthine oxidation method. Results Compared with ICH group, the content of water and TNF-a in brain tissue and the level of serum MMP-9 were significantly decreased, the serum SOD activity was significantly increased in Edaravone-treated group and sham operation group (P<0.05 -0.01). Compared with sham operation group, the difference of brain water content with Edaravone-treated group was significantly (P <0.05). Brain water content of Edaravone-treated group was positive correlated with serum MMP-9 (r = 0. 956, P = 0.003 ) , negative correlated with the activity of SOD (r =-0.945 ,P=0.004 ) . Conclusions Edaravone can decrease the content of water, TNF-a in brain tissue and the level of serum MMP-9, while increase the activity of SOD. Its brain protective effect may through these mechanisms.关键词
