Abstract
The aim of this study was to investigate whether hepatic concentrations and mRNA expression of metallothionein( MT) in broilers are well-accepted biomarkers for the bioavailability of different forms of zinc. Three sources of zinc, graded levels of ZnSO4·H2O,ZnO and ZnAA,were added to the basal diet,respectively. Then hepatic concentrations and mRNA expression of MT in broilers were measured at the weeks of 2,3,4,5 and 6 during the entire experimental period. The results showed that there was significant difference in hepatic MT concentrations and mRNA expression between interspecies, with those of Gushi chick greater than those of AA broilers. Both the sources and levels of zinc affected the experimental parameters(P<0.05). ZnAA exhibited the most, followed by ZnSO4 · H20 and both of them were significantly superior to ZnO. With the increasing levels of zinc, hepatic concentrations and mRNA expression of MT increased, and those were significantly influenced by the zinc levels. For the first 3 weeks,there were pronounced increases in hepatic concentrations and mRNA expression of MT in broilers, whereas both were decreasing with the age increasing since the 4th week. The bioavailability determined by the MT concentrations for various sources of zinc was in accordance with the results derived from the MT mRNA expression, which demonstrated that the concentrations and mRNA expression of hepatic MT were well-defined biomarkers for the bioavailability of zinc. Moreover, compared to the expression of MT mRNA,the MT concentrations were more sensitive to the species,age and the level of zinc.关键词
