Abstract

An Expressed Sequence Tag (EST) contained a MADS conserved sequence was screened from Hevea brasiliensis cDNA subtractive library between early flowering material and common germplasm. Primers were designed based on the EST sequence to amplify the complete sequence using 5' Rapid Amplification of cDNA Ends (RACE) by PCR; and a 935 bp sequence named HbA GL62 was obtained. Bioinformatics analysis showed that HbAGL62 had a 795 bp open reading frame and encoded a protein of 265 amino acids containing a MADS conserved domain. HbAGL62 is closer to AGL62 genes of Arabidopsis thaliana, grape and alfalfa gene in genetic evolution tree. Results of Real-time quantitative PCR showed that the expressing of HbAGL62 in rubber tree had spatial and time specificity, HbAGL62 was expressed not in roots and bark, but in flowers and embryonic; meanwhile, not expressed in the leaf bud differentiation stage, but highly expressed in the flower bud differentiation stage, which indicated that HbAGL62 might be play an important role in flowering regulation of rubber tree.
