荔枝花蕊蛋白双向电泳体系的建立与优化
Establishment and Optimization of Two-dimensional Electrophoresis System for Proteins of Litchi Flower Buds

Abstract
The two-dimensional electrophoresis (2-DE) technology was used to demonstrate the protein profile of Litchi (Litchi chinensis Sonn., cultivar Yuanhong) flower buds. The key processes of sample extraction method, sample loading, gel concentrations, equilibrium time and staining methods of 2-DE system were optimized. Proteins were extracted by phenol extraction method. The results showed that more than 1 200 protein spots were detected by Image Master 2D Platinum with the parameters including pH 4-7 24 cm IPG strip, 1.2 mg protein sample loading, 12% T gel, 20 min equilibrium and Coomassie Brilliant Blue (CCB) R250 stain. The optimized system provided the foundation for further proteomics analysis of flower sex-differentiation in Litchi.关键词
