Abstract

Objective To investigate the effects of testosterone levels on vascular endothelial function in castrated male rats. Methods Thirty male Wistar rats were randomized into 3 groups: rats underwent sham operation (Sham group, n = 10), rats were castrated (Cas group, n = 10) and rats were castrated and supplemented with testosterone 2 mg/kg (TC group 2 d, n = 10). Blood concentrations of testosterone, nitric oxide (NO), endothelin-1 (ET-1) were measured after 8 weeks. Results Compared with the sham group, the plasma testosterone concentration in Cas group was significantly lower (P < 0.05), and that in Tc group was not significantly changed (P > 0.05); plasma NO level in Cas group was significantly lower(P <0.05), and no change in TC group(P > 0. 05); plasma ET-1 content in Cas was significantly higher(P <0. 05), and no significant change in TC group (P > 0.05). Compared with Cas group, plasma testosterone and NO concentrations were significantly increased; whereas plasma ET-1 level was reduced in TC group( P <0.05). Conclusion Change of testosterone levels can affect the vascular endothelial function in castrated male rats. Testosterone deficiency results in vascular endothelial damage, followed by reduced NO and increased ET-1.
